The Dua is mentioned in surah Al Anbiya verse 83.

- And [remember] Ayyoob, when He called upon his Lord: Affliction has indeed befallen me, and You are the Most Merciful of those who show mercy.
- So We answered his prayer and relieved his affliction, and We restored his family to him and doubled their number, as a mercy from Us and as a reminder for all who worship Us.
Explanation from Tafseer Ibn Kathir
He (Ayyub عَلَيْهِ ٱلسَّلَامُ) had plenty of livestock, cattle and crops, many children and beautiful houses, and he was tested in these things, losing everything he had.
Then he was tested with regard to his body, and he was left alone on the edge of the city and there was no one who treated him with compassion apart from his wife, who took care of him.
It was said that it reached the stage where she was in need, so she started to serve people (to earn money) for his sake. The Prophet ﷺ said:
The people who are tested the most severely are the Prophets, then the righteous, then the next best and the next best.
According to another Hadith: A man will be tested according to his level of religious commitment; the stronger his religious commitment, the more severe will be his test.
The Prophet of Allah, Ayyub, upon him be peace, had the utmost patience, and he is the best example of that. Yazid bin Maysarah said:
“When Allah tested Ayyub, upon him be peace, with the loss of his family, wealth and children, and he had nothing left, he started to focus upon the remembrance of Allah, and he said:
`I praise You, the Lord of lords, Who bestowed His kindness upon me and gave me wealth and children, and there was no corner of my heart that was not filled with attachment to these worldly things, then You took all of that away from me and You emptied my heart, and there is nothing to stand between me and You.
If my enemy Iblis knew of this, he would be jealous of me. ‘ When Iblis heard of this, he became upset. And Ayyub, upon him be peace, said:
`O Lord, You gave me wealth and children, and there was no one standing at my door complaining of some wrong I had done to him. You know that. I used to have a bed prepared for me, but I forsook it and said to myself: You were not created to lie on a comfortable bed. I only forsook that for Your sake.”‘
(as a mercy from Ourselves) means, `We did that to him as a mercy from Allah towards him.’
(and a Reminder for all those who worship Us.) means, `We made him an example lest those who are beset by trials think that We do that to them because We do not care for them, so that they may take him as an example of patience in accepting the decrees of Allah and bearing the trials with which He tests His servants as He wills.’ And Allah has the utmost wisdom with regard to that.
The Affliction of Ayyūb عَلَيْهِ ٱلسَّلَامُ lasted 18 years !!
{(Remember) Ayyūb when he cried to his Lord, “I have been stricken by hardship, and You are the Most Merciful of all who show mercy.’} [Sūrah al-Anbiyāʾ 21:83]

Anas (may Allāh be pleased with him) reported that the Prophet ﷺ said: Indeed, Allāh’s prophet Ayyūb remained in his affliction for eighteen years.
Near and far people abandoned him – except for two men from among his brothers who continued to visit him in the morning and evening.“
One day one of these two men said to his companion: ”You should know, by Allāh that Ayyūb must have committed a sin that no person has ever committed.“
His companion asked him: ”Why do you say this?“
He replied: ”Because for eighteen years Allāh has not granted him mercy and removed his affliction.“
When they went to Ayyūb that evening, the second man could not refrain from telling that to Ayyūb.
Ayyūb responded: ”I am not sure to what you two are referring. Surely, Allāh knows that I used to pass by two conflicting men mentioning Allāh (improperly), and I would go back to my house and give a ransom on their behalf, hating that Allāh be mentioned except in a good way.“
When Ayyūb went out to relieve himself, his wife used to wait for him and then hold his hand and help him return home.
One day, she waited and he did not come back. The revelation came then to Ayyūb {Strike the ground with your foot, and this is (the result): a spring for a cool bath and drink} [Ṣad 38:42]
When Ayyūb took too long, his wife went forth looking for him. Then he came toward her after Allāh had alleviated his affliction, and he returned to his original youthful appearance.
When she saw him she asked: ”Excuse me, bārak Allāhu fīk (may Allāh bless you). Have you seen that afflicted Prophet of Allāh? Indeed Allāh being my witness, I have not seen anyone who resembles him – when he was well – like you do.“
He replied: ”I am he!”
{As a reward for his patience} Ayyūb had two large piles of grains for threshing: one of wheat, and one of barley. Allāh sent two clouds. When one of them was over the wheat pile, it poured over it gold until it was overrun with it. The other cloud poured silver over the barley pile until it was overrun with it.

Tafsir And Commentary On These Verses
That is, remember Our slave and Messenger Ayyoob, in a context of praise and respect, showing his high status, when Allah tested him with a great trial and found him patient and content with his Lord.
The Shaytan was given power over his body as a test from Allah, so he breathed into his body and he was afflicted with great sores that stayed for a long time.
He faced a severe calamity, his family died and he lost his wealth, so he called out to his Lord:
O Lord, Affliction has indeed befallen me, and You are the Most Merciful of those who show mercy.
He beseeched Allah by telling of his situation, and that his affliction had become so severe, and by referring to His abundant mercy.
So Allah responded to him and said to him:
…Strike [the ground] with your foot. Here is cool water for you to wash in and drink.
Surah Sad 38: verse 42
So he struck the ground with his foot, and as a result of his doing so, a spring of cold water gushed forth.
He bathed in it and drank from it, and Allah relieved him of his affliction.
And We restored his family to him) that is, we returned his family and his wealth to him and doubled their number) because, in addition to healing him, Allah granted him family and wealth in abundance.
As a mercy from Us) to him, because he was patient and was content with his Lord.
Therefore Allah rewarded him in this world, before bestowing the reward of the hereafter.
And as a reminder for all who worship Us) that is, We made him a lesson for all who worship Us, those who will benefit from the lesson.
When they see what befell him of calamity, then what Allah rewarded him with after it was over, and they look for the reason behind that, they will realise that it is patience and steadfastness.
Hence Allah praised him for that when He said:
.. .Truly We found him to be patient and steadfast, a good and faithful slave who constantly turned [to Us].
Surah Sad 38: 44
So they will take him as an example to follow when calamity strikes.
